A Comparison of SPEAD and BEAD Sample Pre-Treatment to Improve Residual Drug Tolerance in a Cell-Based NAb

EBF Open Symposium 2020 -- Residual drug tolerance issues in cell-based NAbs [Neutralizing Antibody (NAb) assays] is a common problem (particularly for IgG molecules), which can lead to false negative NAb detection. IgG molecules typically circulate at high (�g/mL) levels to achieve efficacy and also have long wash out periods due to recycling by FcRn receptors. Extraction with acid dissociation is a common approach used to address this. Using a cell-based assay system residual drug tolerance was determined and compared for both BEAD (Bead Extraction & Acid Dissociation) & SPEAD (Solid Phase Extraction & Acid Dissociation) platforms.
